产品详细信息It is not recommended to aliquot this antibody.靶标信息NFkB is a nuclear transcription factor activated by various extra and intracellular stimuli such as cytokines, UV radiation, stress, in injury, and by bacterial and viral products. NFkB is involved in regulation of various cellular events including cell growth, differentiation, proliferation, apoptosis and inflammation. NFKB1 (p50) is a 50KDa functional sub-unit of NF-kB and a member of Rel protein family which is synthesized as a p105 precursor protein and consists of an N-terminal conserved RHD-region containing nuclear localization signal, DNA-binding and dimerization domains. NFKB1 (p50) forms homodimers or heterodimerizes with p65, forming the functional NFkB factor. Further, NFKB1 (p50) directs the nuclear translocation of NF-kB and is instrumental in its DNA-binding. The NFkB complex is expressed in most cell types, and is primarily found in the cytoplasm in an inactive state in association with I-kappa-B. Phosphorylation of I-kappa-B releases the complex, allowing translocation to the nucleus and modification of gene expression. Phosphorylation of the p50 subunit at the Ser-337 residue has been shown to significantly increase DNA binding efficiency of the NFkB protein complex. The pathological role of NFkB has been suggested in AIDS, hematogenic cancer cell metastasis and rheumatoid arthritis.

NFkB p50 Antibody (PA5-17654) in WBWestern blot analysis of NFKB1 was performed with 10 µg of A549 cells transfected with Transfection Reagent alone (Lane 1), 100nM Non-Targeting control siRNA (Lane 2), or 100nM siRNA against NFKB1 (Lane 3). Proteins were resolved using a NuPAGE® Novex 4-12% Bis-Tris Gel (Product # NP0322BOX), XCell SureLock™ Electrophoresis System (Product # EI0002), and a protein size ladder. Proteins were wet transferred to a Pierce Nitrocellulose Membrane (Product # 88025) OR Pierce PVDF Membrane (Product # 88518) and blocked with Pierce Starting Block T20 (PBS) Blocking Buffer (Product # 37539) for 1 hour at room temperature. NFKB1 was detected at ~ 105 and 50 kDa using NFKB1 Rabbit polyclonal antibody (Product # PA5-17654) diluted in Pierce Starting Block T20 (PBS) Blocking Buffer 4°C overnight on a rocking platform. Pierce Goat Anti-Rabbit (Product # 31461) HRP-Conjugated Antibodies at a 1:2500 dilution were used and chemiluminescent detection was performed using Pierce Supersignal West Dura Maximum Sensitivity Substrate (Product # 37071). Relative density of the bands normalized to GAPDH (36 kDa). NFKB1 Antibody (Product # PA5-17654) confirms silencing of NFKB1 expression.
