产品详细信息This antibody is predicted to react with Rat, Bat, Cat and PigRecombinant rabbit monoclonal antibodies are produced using in vitro expression systems. The expression systems are developed by cloning in the specific antibody DNA sequences from immunoreactive rabbits. Then, individual clones are screened to select the best candidates for production. The advantages of using recombinant rabbit monoclonal antibodies include: better specificity and sensitivity, lot-to-lot consistency, animal origin-free formulations, and broader immunoreactivity to diverse targets due to larger rabbit immune repertoire.靶标信息Histone H3 is one of the DNA-binding proteins found in the chromatin of all eukaryotic cells. H3 along with four core histone proteins binds to DNA forming the structure of the nucleosome. Histones play a central role in transcription regulation, DNA repair, DNA replication and chromosomal stability. Post translationally, histones are modified in a variety of ways to either directly change the chromatin structure or allow for the binding of specific transcription factors. The N-terminal tail ofhistone H3 protrudes from the globular nucleosome core and can undergo several different types of post-translational modification that influence cellular processes. These modifications include the covalent attachment of methyl or acetyl groups to lysine and arginine amino acids and the phosphorylation of serine or threonine.

H3K79me2 Antibody (701770) in ICCFor immunofluorescence analysis, HeLa cells were fixed and permeabilized for detection of endogenous H3K79me2 using Histone H3K79me2 Recombinant Rabbit Monoclonal Antibody (Product # 701770, 2 µg/mL) and labeled with Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, Alexa Fluor® 488 conjugate (Product # A27034, 1:2000). Panel a) shows representative cells that were stained for detection and localization of H3K79me2 protein (green), Panel b) is stained for nuclei (blue) using SlowFade® Gold Antifade Mountant with DAPI (Product # S36938). Panel c) represents cytoskeletal F-actin staining using Alexa Fluor® 555 Rhodamine Phalloidin (Product # R415, 1:300). Panel d) is a composite image of Panels a, b and c clearly demonstrating nuclear localization of H3K79me2. Panel e) shows loss of signal by competition with the H3K79me2 peptide, demonstrating antibody specificity, and panel f) demonstrates no competition with the non-methylated peptide. Panel g) represents control cells with no primary antibody to assess background. H3K79me2 Antibody (701770) in ChIPEnrichment of endogenous Di Methyl Histone H3 (Lys 79) (Histone H3K79me2) protein at specific gene loci using Anti- Histone H3K79me2 Recombinant Rabbit Monoclonal Antibody: Chromatin Immunoprecipitation (ChIP) was performed using Anti-Histone H3K79me2 Recombinant Rabbit Monoclonal Antibody (Product # 701770, 3 µg) on sheared chromatin from 2 million HeLa cells using the "MAGnify ChIP system" kit (Product # 49-2024). Normal Rabbit IgG (1 µg) was used as a negative IP control. The purified DNA was analyzed by 7500 Fast qPCR system (Product # 4351106) with optimized PCR primer pairs for the promoter of PABCP-1, GAPDH INTRON, IRF1, used as positive control target and the region of the inactive SAT2 satellite repeat, MYOD, used as negative control target gene. Data is presented as fold enrichment of the antibody signal versus the negative control IgG using the comparative CT method. |
