Thermo Scientific MagJET NGS Cleanup and Size Selection kit, sufficient for 10 preps, is designed for efficient and robust DNA fragment library cleanup and size-selection from a variety of enzymatic reaction mixtures including PCR, ligation, adapter addition and DNA end-repair reaction mixes. The kit utilizes high-capacity MagJET paramagnetic bead technology that ensures efficient recovery of DNA fragment library within a desired fragment length range. The purified DNA fragments are free of any next-generation sequencing workflow inhibiting components, such as sequencing adapters, primer dimers, unincorporated nucleotides, enzymes or salts, and are ready-to-use in downstream NGS applications. No prior DNA fragment library purification is required before the cleanup and size-selection procedures.The kit is suitable for cleanup or size selection of 5 ng to 5 µg of DNA fragment library. MagJET cleanup and adapter removal protocols can easily be automated for higher throughput on automatic platforms such as Thermo Scientific KingFisher Flex and Thermo Scientific KingFisher Duo.

图1.Robust cleanup and size selection from various enzymatic mixtures and buffersFragmentedE. coligenomic DNA (1 µg), spiked in different reaction mixtures and buffers (a-h) typical for next-generation sequencing workflow, was size selected using the MagJET and Supplier kits. Isolated fragment libraries were analyzed on agarose gel. Expected size selection peak areas (330/380/400 bp) are marked with a dotted line.

Varying DNA fragment ranges were selected corresponding different sequencing read lengths. Size selection was performed using MagJET NGS Cleanup and Size Selection Kit. DNA library samples were analyzed on Bioanalyzer 2100™ (Agilent Technologies, Inc.).

图3.Efficient sequencing adapter removalPhysically fragmented E. coli genomic DNA (1 µg) samples and 50 bp adapters (7 µg) were added to ligation mixture followed by cleanup step using MagJET adapter removal protocol. Isolated fragment libraries were analyzed by electrophoresis on an agarose gel.
