产品详细信息Description: The 11B11 antibody reacts with mouse interleukin-4 (IL-4), a 14 kD cytokine. The 11B11 antibody is a neutralizing antibody. Mouse IL-4, also known as BCDF, BCGF-1 and BSF-1, is secreted by activated Th2 and natural killer T cells, and to a lesser extent by Th1 and mast cells. IL-4 is species-specific and promotes proliferation and differentiation of B cells. IL-4, in conjunction with other signals such as CD40, induces Ig isotype switching of B cells and the upregulation of surfaceMHC class II and CD23.Applications Reported: This 11B11 antibody has been reported for use in intracellular staining followed by flow cytometric analysis.Applications Tested: This 11B11 antibody has been pre-titrated and tested by intracellular staining of Th2 polarized and stimulated mouse splenocytes. This can be used at 5 µL (0.06 µg) per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test.Excitation: 488-561 nm; Emission: 578 nm; Laser: Blue Laser, Green Laser, Yellow-Green Laser.Filtration: 0.2 µm post-manufacturing filtered.靶标信息Interleukin-4 (IL-4) is a pleiotropic, immune-modulatory cytokine produced by activated T cells, and serves as a ligand for interleukin 4 receptor. IL-4 has been found to have the broadest range of actions. The interleukin 4 receptor also binds to IL13, which may contribute to many overlapping functions of this cytokine and IL13. STAT6, a signal transducer and activator of transcription, has been shown to play an important role in mediating the immune regulatory signal of IL-4. The IL-4 gene, IL-3, IL-5, IL-13, and CSF2 form a cytokine gene cluster on chromosome 5q. In particular, IL-4 is located in close proximity to IL13 on the chromosome. Further, the IL-4 gene, IL13 and IL5 are found to be coordinated by several long-range regulatory elements over a 120 kilobase range on the chromosome. IL-4 delta 2 is a known splice variant of IL-4.

Naive T helper cells were cultured following Th2 activation condition in the presence of IRE1a inhibitor 4mu8c for 3 days, rested for 2 days, reactivated by coated plate, and analyzed by flow cytometry to detect intra-cellular cytokines IL4, IL5, and IL13 expression. Representative FACS profiles are displayed in the first two columns. The intra-cellular cytokine expression is compared in column 3, with three to seven independent biological replicates. Fourth column: cell culture supernatants from 4mu8c-treated or DMSO-treated Th2 were analyzed by ELISA to measure the cytokine concentration.
