It plays an important role in somatic stem cell maintenance and is studied as a stem cell biomarker. It is a zinc finger transcription factor that is upregulated during development and shown to interact with Oct4 and Sox2. It also forms a complex with Nanog to maintain the pluripotency of Embryonic Stem cells (ESCs). Several current research studies focus on its function asan oncogene and tumor biomarker. Some of the reported target genes of Sall4 include p53, Bmi-1, Bcl2 and XIAP.

SALL4 Antibody (701664) in IFImmunofluorescence was performed on fixed and permeabilized NTERA-2 cells for detection of endogenous Sall4 using Anti-Sall4 Recombinant Rabbit Monoclonal Antibody (Product # 701664, 2 µg/mL) and labeled with Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, Alexa Fluor® 488 conjugate (Product # A27034, 1:2000). Panel a) shows representative cells that were stained for detection and localization of Sall4 protein (green), Panel b) is stained for nuclei (blue) using SlowFade® Gold Antifade Mountant with DAPI (Product # S36938). Panel c) represents cytoskeletal F-actin staining using Alexa Fluor® 555 Rhodamine Phalloidin (Product # R415, 1:300). Panel d) is a composite image of Panels a, b and c clearly demonstrating nuclear localization of Sall4. Panel e) represents control cells with no primary antibody to assess background. The images were captured at 60X magnification. SALL4 Antibody (701664) in ChIPEnrichment of endogenous SALL4 protein at specific gene loci using Anti-SALL4 Recombinant Rabbit Monoclonal Antibody: Chromatin Immunoprecipitation (ChIP) was performed using Anti-SALL4 Recombinant Rabbit Monoclonal Antibody (Product # 701664, 3 µg) on sheared chromatin from 2 million HeLa cells using the MAGnify ChIP system kit (Product # 49-2024). Normal Rabbit IgG was used as a negative IP control. The purified DNA was analyzed by qPCR with PCR primer pairs for the promoter of the HOXA1 positive target gene and HOXA1 Upstream region, GAPDH Promoter used as negative target genes. Data is presented as fold enrichment of the antibody signal versus the negative control IgG using the comparative CT method.
