靶标信息Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. Two molecules of each of the four core histones (H2A, H2B, H3, and H4) form an octamer, around which approximately 146 bp of DNA is wrapped in repeating units, called nucleosomes. The linker histone, H1, interacts with linker DNA between nucleosomes and functions in the compaction of chromatin into higher order structures. This gene is intronless and encodes a member of the histone H4 family. Transcripts from this gene lack polyA tails but instead contain a palindromic termination element. This gene is found in the large histone gene cluster on chromosome 6.

H4K5ac Antibody (PA5-40085) in ICCImmunofluorescence analysis of Acetyl-Histone H3 (Lys18) was performed using 70% confluent log phase HeLa cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 10 minutes, and blocked with 1% BSA for 1 hour at room temperature. The cells were labeled with Acetyl-Histone H4 (Lys5) Rabbit Polyclonal Antibody (Product # PA5-40085) at 1:250 dilution in 0.1% BSA, incubated overnight at 4 degree Celsius and then labeled with Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, Alexa Fluor® 488 conjugate (Product # A27034) at a dilution of 1:2000 for 45 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with SlowFade® Gold Antifade Mountant with DAPI (Product # S36938). F-actin (Panel c: red) was stained with Rhodamine Phalloidin (Product # R415, 1:300). Panel d represents the merged image showing nuclear localization. Panel e shows control cells with no primary antibody to assess background. The images were captured at 60X magnification. H4K5ac Antibody (PA5-40085) in WBWestern blot analysis was performed on acid extracts (20 µg lysate) of HeLa (Lane 1), HeLa treated with sodium butyrate (5mM sodium butyrate for 24 hr) (Lane 2), HCT 116 (Lane 3) and HCT 116 treated with sodium butyrate (5mM sodium butyrate for 24 hr) (Lane 4). The blot was probed with Anti-Acetyl Histone H4 (Lys 5) (Product # PA5-40085, 1:500 dilution) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/mL, 1:4000 dilution). A 14 kDa band corresponding to Acetyl Histone H4 (Lys 5) was observed across the cell lines tested.
