产品详细信息This antibody is predicted to react with equine, rabbit and rat based on sequence homology.Intact IgG appears on a non-reducing gel as ~150 kDa band and upon reduction generating a ~25 kDa light chain band and a ~50 kDa heavy chain.Recombinant rabbit monoclonal antibodies are produced using in vitro expression systems. The expression systems are developed by cloning in the specific antibody DNA sequences from immunoreactive rabbits. Then, individual clones are screened to select the best candidates for production. The advantages of using recombinant rabbit monoclonal antibodies include: better specificity and sensitivity, lot-to-lot consistency, animal origin-free formulations, and broader immunoreactivity to diverse targets due to larger rabbit immune repertoire.靶标信息CDf (Integrin alpha-6, ITGA6) is the integrin alpha chain alpha 6 protein. Integrins are integral cell-surface proteins composed of an alpha chain and a beta chain. A given chain may combine with multiple partners resulting in different integrins. For example, alpha 6 may combine with beta 4 in the integrin referred to as TSP180, or with beta 1 in the integrin VLA-6. Integrins are known to participate in cell adhesion as well as cell-surface mediated signaling. Two transcript variants encoding different isoforms have been found for CDf. Functionally, integrins are cell surface receptors that interact with the extracellular matrix and mediate intracellular signals that affect cellular shape, mobility, and progression through the cell cycle in response to the extracellular matrix. Integrin receptors are composed of alpha and beta subunits, and form structural and functional linkages between the ECM (extra-cellular matrix) and intracellular cytoskeletal linker proteins. Signaling mediated by Intergrin/ECM interactions are also integrated with cellular responses to growth factor signaling to regulate cellular proliferation, cytoskeletal reorganization and other responses necessary for cellular survival. CDf is predominantly expressed by epithelia, and plays a critical structural role in the hemidesmosome. More than 18 alpha and 8 beta subunits with numerous splice variant isoforms have been identified in mammals. Certain integrins can also bind to soluble ligands or to counter-receptors on adjacent cells, such as the intracellular adhesion molecules (ICAMs), resulting in aggregation of cells.

CD49f Antibody (701178) in WBWestern blot analysis was performed on whole cell extracts (30 µg lysate) of U-87 MG (Lane1), MDCK (Lane2), CaCo-2 (Lane 3) and A549 (Lane4). The blots were probed with Integrin alpha 6 Rabbit Monoclonal Antibody (Product # 701178, 2-3 µg/mL) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.4 µg/mL, 1:2500 dilution). A 150 kDa band corresponding to Integrin alpha 6 was observed across cell lines tested. Known quantity of protein samples were electrophoresed using Novex® NuPAGE® 10% Bis-Tris gel (Product # NP0302BOX), XCell SureLock™ Electrophoresis System (Product # EI0002), and Novex® Sharp Pre-Stained Protein Standard (Product # LC5800). Resolved proteins were then transferred onto a nitrocellulose membrane with Pierce™ Power Blotter System (22834) The membrane was probed with the relevant primary and secondary Antibody following blocking with 5 % skimmed milk. Chemiluminescent detection was performed using Pierce™ ECL Western Blotting Substrate (Product # 32106).
