靶标信息Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. Two molecules of each of the four core histones (H2A, H2B, H3, and H4) form an octamer, around which approximately 146 bp of DNA is wrapped in repeating units, called nucleosomes. The linker histone, H1, interacts with linker DNA between nucleosomes and functions in the compaction of chromatin into higher order structures. This gene is intronless and encodes a member of the histone H4 family. Transcripts from this gene lack polyA tails but instead contain a palindromic termination element. This gene is found in the large histone gene cluster on chromosome 6.

H4K20me3 Antibody (PA5-40089) in IFImmunofluorescent detection of Tri-Methyl-Histone H4 (Lys20) in human osteosarcoma (U2OS) cells using a Tri-Methyl-Histone H4 (Lys20) polyclonal antibody (Product # PA5-40089). Cells were fixed with ice cold methanol for 10' and blocked with PBS/Triton X-100 containing 5% normal goat serum. Figure 6A: cells were immunofluorescently labeled with the H4K20me3 antibody (left) at a dilution of 1:300 in blocking solution followed by detection using an anti-rabbit antibody conjugated to AlexaFluor 568 or with DAPI (right), which specifically labels DNA. H4K20me3 Antibody (PA5-40089) in ELISATo determine the titer of the antibody, an ELISA was performed using a serial dilution of the anti-H4K20me3 antibody (Product # PA5-40089), crude serum and flow through in antigen coated wells. The antigen used was a peptide containing the histone modification of interest. By plotting the absorbance against the antibody dilution, the titer of the antibody was estimated to be 1:7,400.
