产品详细信息MA1-045 has been successfully used in immunofluorescence analysis of PSD95 in human motor neurons derived from iPSCs.靶标信息Post Synaptic Density 95 kDa (PSD-95), also known as synapse associated protein 90 kDa (SAP90), is one of a family of membrane-associated proteins found in the postsynaptic density in forebrain neurons and certain presynaptic structures in the cerebellum. Like other members of the family, PSD-95 has three 90 amino acid repeats called PDZ domains followed by an SH3 domain and a yeast guanylate kinase homology (GuK) domain. PSD-95 is believed to participate in the clustering of certain proteins, including NMDA receptors, Shaker-type potassium channels at the synaptic membrane in central nervous system (CNS) neurons. There are two principal modes of interaction between PSD-95 and other proteins. NMDA receptors and shaker-type potassium channels both share C-terminal sequence homology consisting of a threonine/serine-X-valine-COOH (T/SXV) motif. Other neuronal proteins that share this motif (beta 1 adrenergic receptor, some serotonin receptors, some sodium channel subunits, and additional potassium channel subunits), and some of these proteins may interact with PSD-95 by binding to its PDZ domains. Neuronal nitric oxide synthase (nNOS), which lacks the T/SXV motif but which has its own PDZ domain, has been shown to associate with PSD-95 in vitro through a pseudo-homotypic PDZ-PDZ interaction.仅用于科研。不用于诊断过程。未经明确授权不得转售。

PSD-95 Antibody (MA1-045) in IFImmunofluorescent analysis of Post Synaptic Density 95kD protein (PSD95, green) in cultured primary cortical neurons. Primary cortical neurons are isolated and cultured using the Primary Neuron Isolation Kit (Product # 88280). At day 28, neurons were fixed with 4% paraformaldehyde, permeablilized with 0.1% triton X-100 in HBSS for 10 minutes at room temperature, and blocked with 3% BSA in PBS (Product # 37525) for 30 minutes at room temperature. Cells were probed with a PSD95 monoclonal antibody (Product # MA1-045) at a dilution of 1:500 for 2 hours at room temperature or overnight at 4 °C, washed with HBSS, and incubated with DyLight 488 goat anti-mouse IgG secondary antibody (Product # 35502) at dilution of 1:500 for 1 hour at room temperature. Images were taken at 20X magnification on a Carl Zeiss microscopy (AxioVision Rel. 4.7). PSD-95 Antibody (MA1-045) in IFImmunofluorescent analysis of PSD95 using PSD95 Monoclonal antibody (6G6-1C9) (Product # MA1-045) shows staining in U251 glioma cells. PSD95 staining (green), F-Actin staining with Phalloidin (red) and nuclei with DAPI (blue) is shown. Cells were grown on chamber slides and fixed with formaldehyde prior to staining. Cells were probed without (control) or with or an antibody recognizing PSD95 (Product # MA1-045) at a dilution of 1:100-1:200 over night at 4 °C, washed with PBS and incubated with a DyLight-488 conjugated secondary antibody (Product # 35552for GAR,Product # 35503for GAM). Images were taken at 60X magnification. PSD-95 Antibody (MA1-045) in FlowFlow cytometry analysis of PSD95 in Neuro-2a cells compared to an isotype control (blue). Cells were harvested, adjusted to a concentration of 1-5x10^6 cells/mL, fixed with 2% paraformaldehyde and washed with PBS. Cells were blocked with a 2% solution of BSA-PBS for 30 min at room temperature and incubated with a PSD95 monoclonal antibody (Product # MA1-045) at a dilution of 2 µg/test for 60 min at room temperature. Cells were then incubated for 40 min at room temperature in the dark using a Dylight 488-conjugated goat anti-mouse IgG (H+L) secondary antibody and re-suspended in PBS for FACS analysis. |
