Cytokeratins range in molecular weight between 40 kDa- 68 kDa, and an isoelectric pH between 4.9-7.8. The individual human cytokeratins are numbered 1 to 20. The various epithelia in the human body usually express cytokeratins which are not only characteristic of the type of epithelium, but also related to the degree of maturation or differentiation within an epithelium. Cytokeratin subtype expression patterns are used to an increasing extent in the distinction of different types of epithelial malignancies. The cytokeratin antibodies are not only of assistance in the differential diagnosis of tumors using immunohistochemistry on tissue sections, but are also a useful tool in cytopathology and flow cytometric assays. The composition of cytokeratin pairs vary with the epithelial cell type, stage of differentiation, cellular growth environment, and disease state. Many studies have shown the usefulness of keratins as markers in cancer research and tumor diagnosis.

Cytokeratin Pan Type I/II Antibody (MA1-82041) in IFImmunofluorescence analysis of Cytokeratin Pan Type I/II was performed using 70% confluent log phase A-431 cells. The cells were fixed and permeabilized with ice-cold 100% acetone and blocked with 1% BSA for 1 hour at room temperature. The cells were labeled with Cytokeratin Pan Type I/II Monoclonal Antibody (AE1/AE3) (Product # MA1-82041) at 1:100 dilution in 0.1% BSA, incubated at 4 degree Celsius overnight and then labeled with Goat anti-Mouse IgG (H+L) Superclonal™ Secondary Antibody, Alexa Fluor® 488 conjugate (Product # A28175) at a dilution of 1:2000 for 45 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with ProLong™ Diamond Antifade Mountant with DAPI (Product # P36962). F-actin (Panel c: red) was stained with Rhodamine Phalloidin (Product # R415). Panel d represents the merged image showing cytoskeletal localization. The images were captured at 60X magnification. Cytokeratin Pan Type I/II Antibody (MA1-82041) in WBWestern blot analysis of cytokeratin was performed by loading 20 µg of a negative control fibroblast lysate (Lanes 1-2) or a lysate from equine keratinocytes isolated from scrotal skin of a healthy male Thoroughbred horse (Lanes 3-4) per well onto an SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% milk for 1 hour. The membrane was probed with a Pan Cytokeratin monoclonal antibody (Product # MA1-82041) at a dilution of 1:500 for 2 hours at room temperature, washed in TBST, and probed with an HRP-conjugated goat anti-mouse IgG secondary antibody at a dilution of 1:50,000 for 30 minutes.
