Ubiquitously expressed in low levels, PRDM2 exists in 2 isoforms, a 280 kDa RIZ1 containing PR-domain and a 250 kDa truncated RIZ2 that lacks PR-domain. A tumor suppressor with a prominent role in cell growth and differentiation, PRDM2 binds to Retinoblastoma protein, estrogen receptor, GATA-3 and TPA-responsive element (MTE) of Heme-oxygenase-1 gene and influences pathogenesis of Retinoblastoma, transcriptional regulation during neuronal differentiation, transactivation of Heme-oxygenase-1 gene and specific effector of estrogen action. It is inactivated or altered due to mutations and hypermethylation in many types of human cancers suggesting a potential biomarker.
